The synthesis of pteroylglutamic (folic) acid in 1945 not only provided pure substrate to make possible studies on the physiology and metabolism of this vitamin, but also led the way to the synthesis of antifolate compounds. These were among the first drugs to be used in cancer chemotherapy. For some time, studies of folate absorption were virtually limited to studies with synthetic folic acid but gradually other forms of folate including polyglutamyl folates were subjected to analysis. It is only in recent years that studies of methotrexate absorption have been undertaken. It is now clear that the transport of methotrexate and the folates in the intestine is mediated by the same mechanism. In this article on folate and antifolate absorption, a detailed analysis of the studies with folic acid will be presented and this will be followed by an evaluation of the experiments with antifolate substrates.
