METHODSTwo hundred fifty to four hundred million bone marrow cells were prepared from B10. BR mice (Jackson Laboratory, Bar Harbor, Maine) and were injected into lethally irradiated (950 to 1100 cGy) Lewis rats (Harlan Sprague Dawley, Indianapolis, Ind). Peripheral blood lymphocytes of animals surviving for 30 days were typed by ffow cytometric analysis using anti-rat (RT1Aavl-FITC) and antimouse (H2Kk-PE) monoclonal antibodies (Pharmingen, San Diego, Calif) to determine their relative percentages. All animals were monitored for signs of GVHD. Severe diarrhea, loss of hair, dermatitis, and wasting mark the onset of GVHD. In suspect rats histologic evidence of GVHD was sought.
