Tamoxifen (5 mg/day .times. 2) administered s.c. at the time of DMBA [7,12-dimethylbenz(a)anthracene) administration reduced the number of mammary tumors found 120 days later to < 10% of controls. A 2nd dose regimen of tamoxifen 30 days after DMBA inhibited tumor genesis up to 120 days. Tamoxifen (5 mg/day .times. 2) administered to rats with established mammary tumors produced antitumor effects for 3 wk. Treated tumors were significantly (P < 0.01) smaller than controls. The binding of [3H]estradiol-17.beta. in vivo was significantly lower in uteri (P < 0.01) vaginae (P < 0.001) and mammary tumors (P < 0.01) of treated animals compared with controls. Tamoxifen (5 mg/day .times. 2) inhibited estrogen-stimulated elevations in plasma prolactin (P < 0.01) and uterine wet weights (P < 0.001) in ovariectomized rats. Inhibition of estrogen action at the tumors level probably plays a more fundamental role in the antitumor effects of tamoxifen than reduction in circulating prolactin levels.
