Marked differences in the AChE activity of rat motor and sensory spinal roots were consistently observed by means of the histochemical method of Karnovsky. Using 11 ventral roots (VR) and 10 dorsal roots (DR) from the segments L3and L4, we assessed the accuracy with which motor and sensory nerve fibres may be distinguished by histochemical staining. The percentage of stained fibres increased as the incubation period was lengthened. The difference in the percentage of stained fibres between DR and VR was small after short incubations and also after very long incubation periods. The maximal difference was found at about 20–26 h of incubation. This period is called ‘the critical incubation period’ for the differentiation between motor and sensory nerve fibres. The difference never reached 100%. Even after ‘critical incubation’ 17.7% of VR fibres were still unstained, whereas 8.9% of DR fibres were stained. This overlap of staining to some extent reduces the exactness of differentiation between motor and sensory axons.Sympathetic preganglionic fibres show the highest AChE activity of VR fibres, little less activity is present in fusimotor fibres, and still lower activity is characteristic of Aα fibres.
