Operations were done on near-term pregnant rabbits in which the tracheas of half the fetuses were ligated to prevent aspiration of amniotic fluid and sham operations performed on the other half. The fetuses were allowed to remain in utero for varying periods up to 5 days. They were then delivered by cesarean section and the heads tied off before a single breath could be taken by the controls. The lungs were dissected free, lung weights and volumes measured, and histological preparations made. Significant increase of both lung weight and volume was observed in the experimental lungs as compared to those of the sham operated controls. Microscopic examination showed normal alveolar development, but in the experimental lungs there was also evidence of dilatation of alveoli and terminal bronchioles as well as thinning of their walls. It is logical to conclude that a fluid is produced in the lung against a hydrostatic pressure and that normal lung development occurs in the absence of amniotic fluid aspirations.
