The method for preparing viable brain slices that exhibit electrophysiologica [properties similar to some of those found in in-vivostudies has permitted intmcelluhtr recording from human glia. The method is made more powerful by iontophoresing a marker (eg horseradish peroxidase) through the recording electrode to obtain positive morphological identification of the cell being studied. Accumulating results demonstrate the potential of this method for gaining new information about normal glial function and the pathophysiology of epilepsy and glial tumors in the haman brain.In the mammalian brain, gila outnumber neurons ten to one, In general the larger the brain the greater the ratio of gila to neurons, suggesting that gila should have an important function. Yet their physiological significance has eluded neurobiologists and speculation about their function has ranged from considering them to be merely a'nerve cement'(neuroglia), to having an important role in learning and memoryL The elegant studies by Kuffier and coworkers 5, demonstrating that gila have an exclusive permeability to K'in the CNS of the invertebrate Icech, provided the first quantitative physiological data about gila. This discovery, and the applicability of recently introduced techniques (eg tracer methods, immunological, enzymological, freeze fracture) to the study of glia are responsible for the burgeoning interest in these cells.
