We have recently demonstrated that the rat corpus luteum synthesizes both insulin-like growth factor I (IGF-I) and IGF-I receptor mRNAs (1). IGF-I is a mitogenic factor known for its stimulatory effect on cell proliferation (2). In certain cells and specifically rat granulosa cells, IGF-I also causes cellular differentiation and maturation. Therefore, in the granulosa cells, IGF-I may have a dual function as modulator of growth and differentiation (3). The primary culture of rat luteal cells and their long-term maintenance have been difficult. Low cellular yields have limited the possibilities for the study of gene regulation in luteal cells. In order to circumvent the shortcomings of primary cultures, various attempts have been made to immortalize epithelial cells from normal tissues and cell cultures that will retain a spectrum of tissue-specific traits and function. Recently, we have been able to characterize a temperature-sensitive rat granulosa cell line (4) that subsequently allowed the cloning of the rat IGF-I receptor cDNA (5). The goal of this investigation was to immortalize luteal cells and establish a cell line that expresses IGF-I and IGF-I receptors in order to study their respective gene regulation.
