本研究结合Shirasu多孔玻璃(SPG)膜乳化技术和一种新型热凝胶化方法,制备了均匀粒径的ph敏感壳聚糖季铵化微球。本工艺以季铵化壳聚糖溶液与α-β-甘油磷酸酯(α-β-GP)的混合物为水相,通过SPG膜乳化技术分散在油相中,形成均匀的W/O乳液。通过热凝胶法,液滴在37℃固化成微球。整个过程简单而温和。考察了工艺条件对制备的微球性能的影响,得出了最佳制备条件。所得微球直径的变异系数(cv)在15%以下。所得微球具有多孔结构,具有明显的ph敏感性。在酸性溶液(pH 5)中溶解快,在中性溶液(pH 7.4)中保持稳定。微球的ph敏感性也影响其药物释放行为。将牛血清白蛋白(BSA)包封在微球中,在酸性溶液中快速释放,在中性介质中缓慢释放。新型季铵化壳聚糖微球具有ph敏感性,可作为生物医学领域的药物传递系统,如肿瘤靶向药物载体。
In this study, uniform-sized pH-sensitive quaternized chitosan microsphere was prepared by combining Shirasu porous glass (SPG) membrane emulsification technique and a novel thermal-gelation method. In this preparation process, the mixture of quaternized chitosan solution and α-β-glycerophosphate (α-β-GP) was used as water phase and dispersed in oil phase to form uniform W/O emulsion by SPG membrane emulsification technique. The droplets solidified into microspheres at 37°C by thermal-gelation method. The whole process was simple and mild. The influence of process conditions on the property of prepared microspheres was investigated and the optimized preparation condition was obtained. As a result, the coefficient of variation (C.V.) of obtained microspheres diameters was below 15%. The obtained microsphere had porous structure and showed apparent pH-sensitivity. It dissolved rapidly in acid solution (pH 5) and kept stable in neutral solution (pH 7.4). The pH-sensitivity of microspheres also affected its drug release behavior. Bovine serum albumin (BSA) as a model drug was encapsulated in microspheres, and it was released rapidly in acid solution and slowly in neutral medium. The novel quaternized chitosan microspheres with pH-sensitivity can be used as drug delivery system in the biomedical field, such as tumor-targeted drug carrier.
