Oestrogens influence many developmental and physiological responses in target cells by regulating specific gene activity. Their action is mediated by a soluble intracellular receptor that functions as a ligand-dependent transcription factor (I, 2). In addition to their primary role to stimulate or repress rates of specific gene transcription they may also regulate post-transcriptional events and other non-genomic events. In the fourth Mortyn Jones lecture I describe our work on the structure and function of the oestrogen receptor (ER), particularly as it relates to the role of oestradiol and the mechanism by which antioestrogens block oestrogen action. The ER is a member of a large superfamily of transcription factors whose activity depends on the binding of a hormonal ligand. In common with other members of the nuclear hormone receptor family the ER functions from sites in the vicinity of responsive genes, termed response elements, with the properties of transcriptional enhancers (1, 3). Nuclear hormone receptors share two characteristic features, namely a DNA binding domain containing a zinc finger motif and a hydrophobic C-terminal region responsible for ligand binding (3, 4). The family of proteins, currently approximately fifty, includes not only the steroid hormone receptors but also the thyroid hormone and retinoic acid receptors and many others, referred to as orphan receptors, for which a ligand has not been identified (5).
