LncRNA HOX transcriptional antisense RNA, HOTAIR, is a long noncoding RNA composed of 2158 nucleotides. Although it does not encode protein, it plays an important role in gene regulation by modifying chromatin structure. It has been shown that HOTAIR is associated with a variety of human cancers, and its high expression promotes the occurrence and development of tumor, and is related to the poor prognosis of cancer and chemotherapy resistance. The mutual interaction of HOTAIR with microRNAs including miR-34a, miR-454-3p, miR-197, miR-138, miR-203, is participated in tumorigenesis. Metabolic reprogramming is one of the ten major characteristics of tumor, and converting the mode of tumor metabolism has become a new target for cancer therapy. There is increasing attention to the role of HOTAIR in tumor metabolic reprogramming, it can influence tumorous glycometabolism, glutamine metabolism, mitochondrial metabolism through the regulation of GLUT1, mTOR, SirT1, HK2, GLS, UQCRQ. Understanding the specific mechanisms of HOTAIR in tumor metabolic reprogramming contributes to the understanding the mechanisms of tumorigenesis and development and provides a new direction for cancer screening, diagnosis, treatment and prevention.
