Pregnancy in the rabbit can be interrupted by daily injection of 5 pg of 17-P-estradiol benzoate during the first three days after coitus. Such treatment reduces the amount of mucin deposited around the ova while they are in the tubes and so, presumably, the lack of mucin is the factor responsible for the death of the blastocysts soon after they enter the uterus (Greenwald, ’57). This layer which invests the ova has long been referred to as the “albumen layer,” but recently it has been shown to consist primarily of a mucopolysaccharide (Braden, ’52 ; Bacsich and Hamilton, ’54). Other investigators have described the secretion of this layer by the tubal epithelium (Moreaux, ’13; Cohnen, ’27; Westman, ’30) but no one has yet determined the role of hormones in regulating this activity. Accordingly, the writer has attacked the problem in three ways: first, by injecting the most potent natural estrogen (estradiol) into rabbits and comparing the tubal epithelium of these animals with that of controls (in preceding experiments, attention had been centered on the ova) ; second, by attempting to separate the hormones regulating the synthesis of mucin from those concerned with its release from the cell (this was accomplished
