Surface plasmon resonance detection makes it possible to directly measure the binding of an analyte in solution to a surface immobilized binding partner. The immobilization of one binding partner, the possible depletion of analyte close to the sensor surface, and the assumed interaction model are important in the interpretation of binding data. We describe some of the possible effects of immobilization, kinetic analysis on surfaces with low binding capacity, concentration analysis on surfaces with high binding capacity, and how deviations from a 1:1 interaction model may be identified from binding data.
