Piperoxane (20 mg/kg) and yohimbine (1 mg/kg) blocked the hypotensive and respiratory actions of clonidine (5 μg/kg) in chloralosed rats. These doses of piperoxane and yohimbine did not block or reduce clonidine-induced increases in flexor reflex activity, which is dependent on noradrenaline (NA) receptor activity. High doses of piperoxane (20 mg/kg) increased brain NA turnover, and low doses (5 mg/kg) selectively increased cortical NA turnover; similar results occurred after yohimbine. The present results can be explained on the basis of low doses of clonidine selectively stimulating adrenaline (A) receptors which mediate the hypotensive and respiratory actions of clonidine. Adrenaline neurons may, thus, be involved in vasomotor and respiratory control.
