The Bodansky, King-Armstrong, Bessey-Lowry and Klein assay systems for serum alkaline phosphatase were evaluated. The 95% confidence limit of the determination calculated from double-blind duplicate determinations for the first 3 methods ranged from 50% greater to 33% lower than the value obtained. The Klein variability was greater, especially at low values. The first 3 methods were also linearly related to each other over a wide range of phosphatase activity. The Klein method showed a falloff when serums with high alkaline levels were studied. The 4 methods were equally sensitive in detecting a wide variety of clinical diseases.
