Fe-hematoporphyrin compound shows on reduction an absorption spectrum like reduced hematin, and on addition of globin, pyridine or ammonia, gave hemo-chromogen. On addition of a pyridine solution, 0.0316 N in terms of N, to a hematin solution, 0.0316 N in terms of Fe, it was found that in the presence of insufficient pyridine a reduced hematin spectrum is seen together with a pyridine-hemochromogen spectrum. When 2 equivalents of pyridine are added only the hemochro-mogen spectrum is obtained, and further addition of pyridine causes no change. The observed evolution of CO from the CO compound of reduced hematin agreed with the theoretical calculated for 1 molecule of CO in combination with the hematin, both in the absence and presence of pyridine. Solutions of hematin containing different amounts of pyridine were reduced in the presence of CO and the position of the [alpha] band measured. With a limited amount of CO available the a band is first shifted to the red, a minimum reading being obtained when 1 equivalent of pyridine has been added. With further additions of pyridine the [alpha] band again shifts to the violet. With unlimited supply of CO the band shifts to the red, and then remains fairly constant. The author proposes formulae for CO hemo-chromogen and hemocnromogen. He regards reduced hematin as the pigment of hemoglobin shorn of accessory substances, and corresponding to the "haem" of Anson and Mirsky.
