The human TCF7L2 gene was cloned from the CEPH (Centre d'Etude du Polymorphisme Humain) genomic BAC (Bacterial Artificial Chromosome) library. It was shown by fluorescence in situ hybridization (FISH) that it mapped to chromosome band 10q25. 3. This human BAC was used as a probe in FISH experiments to high resolution metaphase chromosomes obtained from PHA-stimulated blood cell cultures of a healthy male. The hybridization signal appeared as yellow spots (FITC) on red chromosomes (counterstained with propidium iodide and DAPI allowing the banding recognition).
