Elution behavior of sodium poly(styrenesulfonate) (NaPSS) compounds on several commercially available columns was studied at varying ionic strengths of phosphate buffer at pH 8.0. A procedure to elucidate the retention behavior of ionic polymers is proposed, which consists of construction of universal calibration curves for both ionic and nonionic polymers at various ionic strengths, comparison of chromatograms of ionic polymers at various ionic strengths, plotting the divergence of universal calibration curves for both ionic and nonionic polymers, and ion-exclusion volume as a function of ionic strength. Universal calibration curves of NaPSS on porous glass columns converged to that of pullulan, but those on other columns went over the pullulan curve with the ionic strength. Ion exclusion is the only secondary effect on porous glass columns and this effect is suppressed at high ionic strength, achieving ideal size exclusion chromatography (SEC). On other columns tested, early elution of NaPSS is governed by ion-exclusion effect, and with increasing the ionic strength, hydrophobic interactions between NaPSS and substrates become remarkable. Retention volume of NaPSS is governed by the compromise among three effects: size-exclusion, ion-exclusion, and hydrophobic interactions. Ideal SEC cannot be expected.
