The localization of histidine-rich protein in the epidermis of developing rat was studied histochemically and immunohistochemically. The findings with the Pauly reaction were identical to those with antisera to histidine-rich proteins having two different molecular weights, both purified from newborn rat epidermis. Histidine-rich protein was detected diffusely throughout the epidermis before fetal granules were formed. Both fetal granules and keratohyalin granules showed concentration of histidine-rich protein(s), but the cells with fetal granules remained at the superficial layer of the body, whereas cells with keratohyalin granules differentiated to form cornified cells. The findings suggest that histidine-rich protein may be chemically heterogeneous and its biological function may not be limited to formation of the matrix in cornified cells.
