A method is described for liberating and estimating intracellular bacteriophage in Escherichia coli at any stage during the latent period. This is brought about by placing the infected bacteria in a medium containing 0.01 [image] cyanide and a high titer T6 lysate. During the 1st half of the latent period both cyanide and T6 are necessary but in the 2d half cyanide alone will bring about lysis and phage liberation. During the 1st half of the latent period no infectious phage is found inside the cells. During the 2d half mature phage accumulates within the bacteria at a rate which is approx. linear and parallel to the increase in intracellular DNA, but lagging several min. behind DNA synthesis. Using 5-methyl-tryptophane in place of cyanide as the metabolic inhibitor similar results are found, but with the intracellular phage curves displaced to the left by several minutes on the time axis.
