METHODSNuclear transfer and embryo culture. Nuclear transfer procedures have been described in detail elsewhere (McGrath and SoRer 1983a, b). Briefly, fertilized ova from spontaneously ovulating females were exposed to 5# g/ml of cytochalasin B (Sigma) and 0.1# g/ml of demecolcine (Sigma) in Whitten's medium (Whitten 1971). Removal of one or both pronuclei within small, plasma membrane-bound, cytoplasmic vesicles was achieved without penetrating the plasma membrane of the egg. The resulting karyoplast was then fused with a previously enucleated zygote by placing it under the zona pellucida together with a small amount of inactivated Sendai virus. These virus-fused nuclear transfer embryos were cultured in Whitten's medium containing 100/~ M EDTA (Abramczuk et al. 1977) or transferred to the oviducts of pseudopregnant foster mothers.
