Mycobacterium tuberculosis strains Erdman, H37Rv, and H37Ra, as well as M. bovis (BCG Tice), were grown on Tween®-albumin, Proskauer and Beck, or American Trudeau Society media. Inocula of the virulent organisms grown on American Trudeau Society and Proskauer and Beck media had a one hundred to one thousand fold lower median lethal dose for mice than the corresponding bacteria grown on Tween-albumin. This correlated with the growth rates determined for the different organisms by the organ enumeration technique. The attenuated organisms showed little or no effect ascribable to the culture medium. Comparisons of the virulence of organisms grown on Proskauer and Beck medium for mice and guinea pigs indicated that the organ involvement index in subcutaneously infected guinea pigs and the initial growth rate in the lungs, livers, and spleens of intravenously infected mice gave comparable assessments of virulence.
