It is well known that calcium ions (Ca") play important roles in cellular functions., such as Inuscle contraction, secretion, and controllingenzylne activities and menlbrane potentials. Hence, scientists have Ineasured intracellular Ca2+ concentrations ([Ca2+]) in cells. To date, severallnethods have been developed in order to Ineasure [Ca+], which include ion-selective electrodes, Ca... indicator dyes, and biosensors. Alnongst them, it is now most popular to use Ca+ indicator dyes such as fura-2, which were developed in the early 1980s (Tsien et ol.t 1982), because application of acetoxy methyl forms of Ca2+ indicator dyes makes loading of the dyes to cells much easier, and because Ca+ imaging is made possible by using Ca+ indicator dyes. The development ofCa2~. indicatordyes is indeed one of the big steps in biology. However, these Ca2+ indicator dyes occasionally cannot satisfy some particular scientific needs. For exanlple, it is not easy to target Ca2+ indicator dyes to specific intracellular organelles, orconversely, unexpected accumulations ofthe dyes to cells and organelles are occasionally observed. In addition, Ca2+ indicator dyes can be extruded from cells. Finally, cell-type specific loadings of Ca+ indicator dyes are not possible, except for some special cases. Since green fluorescent protein (OFP) was cloned from the jellyfish Aequorea victoria (Prasher et a!., 1992), it has been applied intensively in the biological field as a marker or a reporter protein. In addition, it is now possible to make several different types of Ca2+ probes (biosensors) containing GFP.
