Steroidogenesis in Y-l adrenal tumor and 1–10 Leydig tumor cells is sensitive to polylysines of a wide molecular weight range. At low concentrations of polymer there is a 5–10-fold stimulation of steroidogenesis in both cell lines. This is abolished at higher polylysine concentrations. With degrees of polymerization >9–18, half-maximal stimulation occurs at ∼1 × 10−4M monomer concentration irrespective of molecular weight. Adenylate cyclase in partially purified membranes from Y-l and 1–10 cells is stimulated by polylysines which act as partial agonists under these conditions (in contrast to steroidogenesis where responses were comparable to maximal effects obtained with ACTH1–24 or adenosine, respectively). Half-maximal stimulation for all molecular weights of polylysine occurred at 5 × 10−4M when the level is expressed as the concentration of monomer.
