Oncolytic viruses exploit molecular differences between normal and cancer cells to selectively kill the latter. Results of a synthetic lethal screen described in this issue ofCancer Celldemonstrate that components of the unfolded protein response (UPR) limit virus-induced tumor cell killing and identify a strategy to utilize this knowledge.
