The acute phase reactant serum amyloid A (SAA]' is normally a soluble component of high density lipoprotein? Chronic inflammation may result in extracellular deposition of amyloid A (AA) as insoluble fibrils which are thought to be derived from S A A by proteolysis.3 Amyloid P-component (AP], a protein found in amyloid laden tissue, is immunologically indistinguishable from serum amyloid P (SAP), an acute phase reactant in mice? The purpose of this study is to relate the formation of AA fibrils and AP deposition to the acute phase SAA and SAP responses. We have studied the effect of amyloid enhancing factor (AEF] on silver nitrate (AgNO,) induced acute phase SAA and SAP responses. AEF is a substance extracted with 4 M glycerol, .01M Tris, pH 8.5 from pre-amyloidotic spleens of casein treated mice. Kisilevsky and colleaguesS have defined this model of accelerated amyloidosis in which amyloid deposition occurs 48 hours after a subcutaneous dose of 0.5 ml of 2% AgNO, followed by an intravenous injection of AEF. The concentrations of SAA and SAP in serum, and AA and AP in spleen. were measured by solid phase radioimmunoassay (RIA) according to the method of Sipe et a1.6 Spleen tissue was treated with formic acid to solubilize AA fibrils. All spleens were examined by polarization microscopy after Congo Red staining. AgNO, alone produced a large and prolonged SAA and SAP elevation in CBA/J mice (FIGURE 1, top panel], but did not result in AA or AP deposition in spleen. Baseline SAA and SAP concentrations in spleen were 3 ng/mg and 2.3 ng/mg respectively. The maximum acute phase SAA and SAP/AP levels in spleen were 15.0 and 8.4 ng/mg at 48 hours, and had returned to baseline values at 120 hours. No splenic amyloid was detected histologically. When AEF, extracted from whole spleen and cell-free interstitium, was administered simultaneously with AgNO,, splenic AA levels were 250-1200 and 700-900 ng/mg respectively at 48 hours, and amyloid was detected histologically.
