Diet and nutrition are widely recognized to be modulating factors in the metabolism of drugs. This topic has been discussed in several recent reviews (Bidlack et al., 1986; Hathcock, 1985; Anderson et al., 1985; Guengerich, 1984). Dietary factors can affect the absorption and distribution of drugs in the body as well as the'phase I'and'phase II'metabolism of these compounds. Oxygenation is by far the most common pathway in'phase I'metabolism and the reaction is catalyzed by a cytochrome P-450-dependent (P-450-dependent) enzyme system generally known as mixed-function oxidase (MFO). In this review, we shall discuss how diet affects this MFO system and drug metabolism. The MFO enzymes are also important in the metabolic activation of carcinogens and environmental chemicals, and therefore such reactions are included in this article although these toxic chemicals are not drugs. Because of the complexity of the topic, this article is not intended to be an exhaustive review of the existing literature. Instead, we plan to concentrate on the molecular basis of the effects of dietary factors on drug metabolism. Through such analysis, we hope to delineate several concepts that will be useful for interpreting existing results and for planning future experiments in order to gain a better understanding of this topic.
