A simple baiting technique is described which enables the isolation ofPhytophthora megaspermaf. sp.glycineafrom soil with minimal contamination byPythium.The technique involves a preliminary incubation of the soil sample to enable the production of sporangia byPhytophthora in situbefore baiting, followed by a very brief baiting period to minimisePythiumcontamination. The soil sample is flooded to a depth of 1-2 cm above the soil surface, and soyabean leaf disc baits are floated on the water for 1.5 h. Discs are subsequently plated on VYS/PBNRH medium.ADDITIONAL ABSTRACT:Incubation of moistened soil in sealed plastic bags at 21-25 deg C for 1-2 wk provided conditions suitable for production, but not germination, of sporangia. A sample of soil, in a beaker, was then flooded and baited for a brief period only (1-2 h) using soyabean leaf disks.
