A new method for the conversion of flavoproteins to the semiquinoid form is described. This consists of anaerobic photoirradiation in the presence of EDTA [ethylenediamine tetraacetate]. A survey of a number of simple flavoproteins with this technique leads to the conclusion that there exist 2 different classes of flavoprotein free radicals, which are very readily distinguished on the basis of their optical absorption properties. With one enzyme, glucose oxidase, both types of radical are found, the concentrations depending on the pH. These results suggest that the 2 different spectral species may be due to the neutral ?nd anionic radical forms of the flavin coenzyme prosthetic groups.
