Methods are described for the estimation of protein turnover in E. coli by the use of strains with specific amino acid requirements. In rapidly growing cells there was no detectable turnover of protein; in non-growing cells degradation and resynthesis of protein proceeded at the rate of 4 to 5%/hour for several hours. Chloramphenicol, azide and ammonium salts inhibited protein breakdown after a latent period. The effect of 2,4-dinitrophenol was variable. Protein turnover could not be attributed to lysis of cells. The possible function of protein turnover in bacterial metabolism is discussed.
