Protoplasts isolated from mesophyll cells ofDianthus chinensisL. andDianthus caryophyllusL. were fused by polyethylene glycol. Prior to fusion treatment, protoplasts ofD. chinensiswere inactivated with iodoacetamide to inhibit cell division. Protoplasts ofD. caryophyllusdivided to form callus under the culture conditions used in this study but no shoot regeneration was observed. Therefore, fusion-derived calli which regenerated shoots could be tentatively selected as somatic hybrid cell lines. Plants were regenerated from four cell lines, and three of them exhibited intermediate characteristics of both parents in plant height and flower morphology. Esterase isozyme analyses and chromosome counts indicated that they were interspecific somatic hybrid plants. The hybrid nature of these plants was also confirmed by random amplified polymorphic DNA analysis.
