The reversible Na transport blocker amiloride caused current fluctuations at the apical membrane of the outer stratum granulosum of frog skin. Their power density spectra revealed that single transport sites translocate more than 106 Na+/s, which indicated a pore mechanism. The density of open plus amiloride-blocked pores was in the order of 108 pores/cm2 of skin with 60 mM Na and 18 .mu.M amiloride in the outer solution.
