Thrombospodin is a high molecular weight glycoprotein with the ability to bind to heparin,'.' fibr~nectin,'.~ fibrin~gen,"~ plasminogen,6 histidine-rich glycoprotein,' type V collagen,' laminin,' c a l c i~m,~ and sulfated glycolipids." Although the functional significance of these interactions is not known, some of them probably result in the incorporation of thrombospondin into extracellular Thrombospondin has been shown to be present in the extracellular matrices produced by endothelial cells, smooth muscle cells, and fibroblasts in c~ l tu re .~ ' ' ~ McKeown-Longo et aL13 have shown that some of the radiolabeled thrombospondin that is added to human embryonic lung fibroblasts in culture is incorporated into the extracellular matrix. In contrast to fibronectin, the matrix-bound thrombospondin is slowly degraded over a period of hours.I3 Recent studies indicate that thrombospondin biosynthesis by cells in culture is proportional to cell density and is modulated by platelet-derived growth factor and glycosaminoglycans."*16 In addition, the incorporation of thrombospondin into the extracellular matrix can be detected in cultures of human endothelial cells from neonatal skin, but not from adult skin." These data suggest that thrombospondin may have a specific role in the formation of the extracellular matrix and may be affected by variations in biosynthetic phenotype. The developing fibrin clot may represent a specialized example of an extracellular matrix. The incorporation of thrombospondin into fibrin and plasma clots can be demonstrated by determining the amount of radiolabeled thrombospondin that is associated with these clots and by immunofl~orescence.~~~~' Studies of thrombospondin distribution in the clot as a function of time suggest that the majority of thrombospondin is only transiently associated with the clot." Bale et al. I' have reported that thrombospondin is incorporated into the clot while the fibrin is forming. Tuszynski et have reported that more thrombospondin is incorporated into fibrin clots if cross-linking by factor XIII, is inhibited. Thrombospondin is covalently incorporated into high molecular weight complexes in the presence of factor XIII., suggesting that
