METHODSDogs (30-35 kg) of either sex were used. The dogs were anesthetized with pentobarbital sodium (30 mg/kg iv), intubated, and ventilated with oxygen on a Harvard respirator (Harvard Apparatus, Millis, MA). A left thoracotomy was performed, the pericardium was opened, and the proximal left anterior descending coronary artery (LAD) was isolated. The proximal LAD was ligated approximately one-third to one-half the distance from its origin to the apex and distal to the first diagonal branch so as to create a region of intense cyanosis involving approximately 50% of the anterior left ventricle (LV). Dogs were subjected to coronary occlusion for 15 min (n= 3), 1 h (n= 14), or 24 h (n= 6). In these dogs the ligature ‘was released, 5-15 mCi*“Tc-PPi was injected, and reflow was allowed for 1.5 h. An additional group of dogs (n= 4) had 15 min of permanent coronary occlusion without reperfusion. After the dogs were killed, the heart was removed and the LV ischemic tissue (the cyanotic region)(13) and control tissue were divided quickly into subendocardial and subepicardial halves in ice-cold buffer [“buffer I”: 220 mM mannitol, 70 mM sucrose,
