Materials and methodsMost patients entering the trial had tumours refractory to conventional chemotfierapy (Table 4). Thirty-eight patients were entered into this trial (Karnofsky range: 60-90%; median age 52 years). IF together with mesna (dose-relationship 1: 1 on a mg] kg basis) was administered iv continuously over 5 days. The dail. y dose was split into 4 fractions and each fraction was infused over 6 h (Figure 1). The starting dose was 60 mg] kg] 24 h× 5. In Table 1 the different doses of IF are listed and relate to the numffer of patients treated as well as to the treatment cycles.The pharmacokinetics of the cytocidal metabolite 4-hydl: oxy-ifosfamide and the urinary excretion of mesna and active thiol compounds were determined in a further 6 patients. The free sulfhydryl compounds and the thiol compounds, detectable by reduction of the bisulfide bonds, were assayed photometrically at 405 nm by a modified method developed by Ellman (2). 4-hydroxy-ifosfamide was determined fluorometrically (3).
