METHODSGeneral techniques. Experiments were performed on male Sprague-Dawley rats weighing 170-270 g. The rats were anesthetized with an intraperitoneal injection of Inactin (100 mg/kg), and their body temperature was maintained at 37 C by a heated table. A tracheostomy was performed, femoral arterial blood pressure monitored, and maintenance infusion of Ringer bicarbonate solution infused into the femoral vein at 1.5 ml/h. Arterial blood pH and PCO~ were measured on 0.25 ml samples using a Corning blood gas analyzer. The left kidney was exposed using a flank incision, immobilized in a Lucite cup, and surrounded with 3% agar in saline. The kidney surface was bathed with mineral oil equilibrated with 5% CO,/95% 02 and heated to 37 C. Proximal tubule transit times were measured by injecting 0.02 ml of 10% lissamine green intravenously. Only those kidneys whose transit times were less than 12 s were used.
