Recent studies have shown that activated T cells possess cytoplasmic granules containing a set/he proteinase In this article Michael Kramer and Mad (us Simon suggest that the regulated release of this enzyme has an important part to play in the activities of T cells, such as B-cell regulation, cyto~ is and migrationT lymphocytes are induced by structures such as viruses, bacteria, parasites and tumours via specific membraneassociated receptors to express a variety of effector mechanisms. A~ ivated T cells can kill target cells and provide positive or negative signals that regulate cellular and humoral immune responses. There is good evidence that all three T-cell-mediated activities-cytolysis, h~ lp and suppression-are associated with regulated secretion of functional molecules. A multitude of iymphokines are released by activated T helper (]'-h) cells and/or cytolytic T lymphocytes (CTL) such as interleukin 2 (IL-2) 1, interferon gamma (IFN-It) 2, B-cell growth and differentiation factors 3, lymphotoxin 4 and cytolysin/perforin 5.6. However, T cells may secrete and use additional structures-including enzymes-to exert their functions. It has been suggested that T-cell-associated proteinases play an important role in cell killing by CTL 7.8. This assumption was supported by (i) the finding that CTL-mediated and natural killer (NK) cell-mediated target cell lysis is inhibited by low and high molecular proteinase inhibitorsT-l; and (ii) by isolation of a cytolytic serine proteinase from an unselected population of human peripheral blood leukocytes 11. Several recent findings have indicated that proteinases may also participate in T-cell-mediated regulation of humoral immune functions 12-1s. A T-cell derived trypsin like proteinase was shown to be involved in IgE responses 14. B cells can be polyclonaHy activated by various exogenous serine proteinases 12.13 and this process is suppressed by proteinase inhibitors 4. Finally, the fact that proteolytic enzymes-and serine proteinases in particular-are able to degrade basement membrane-like extracellular matrices 16.17 raises the possibility that these enzymatic activities may facilitate the migration of T cells into inflamed tissue.
