One-day-old chicks were fed a calcium-deficient diet (0.43% Ca, 0.3% P) for three weeks and were killed in groups of five at intervals up to 24 hours after an oral dose of 1 mu g 1 alpha ,25-dihydroxycholecalciferol and 100 mu Ci of tritiated thymidine i/p. Duodenal tissue was fixed for radioautography and immunohistological location of calcium-binding protein (CBP). CBP was first seen in the nuclei and cytoplasm of intestinal absorptive cells at the villus bases between 6 and 8 hours and throughout the villus by 18 hours after treatment. Thymidine labelling showed that the progression of CBP appearance from villus base to tip was much faster than cell migration rate. It was concluded that young cells at the base of the villus and more mature cells at the villus tip could synthesize CBP, but that young cells responded faster. The relative intracellular concentration of CBP decreased with time, which suggests that the protein is secreted, degraded by the absorptive cells, or is redistributed in the cells.
