Bulk isolation of immune response-related genes by expressed sequenced tags of Japanese flounder Paralichthys olivaceus leucocytes stimulated with Con A/PMA

In the study, expressed sequence tag sequencing has been coupled with subtractive hybridization to screen for biodefence or immune response-related genes from Japanese flounder (P. olivaceus) peripheral blood leukocytes after treatment with Concanavalin (Con) A/phorbol myristate acetate (PMA) as an in vitro activation system. Database searches identified 43 putative immune response-related genes that were expressed following Con A/PMA stimulation. These putative immune response-related genes are classified into 6 groups: apoptosis-related genes (3 genes), cytokines (7 genes), cytokine receptors (3 genes), signal transduction factors (5 genes), transcription factors (11 genes), and others (14 genes). Results show that fish leukocytes are simulated by, and undergo proliferation following stimulation by mammalian mitogens. During proliferation, they express and utilize various cytokine genes or immune response related genes for communication or signal transduction between cells or intracellular signalling cascades.
