Perhaps an even greater deficiency in many previous reports of cell numbers is the expression of cell number relative to a unit volume of tissue (cell numerical density). It is now well established that components of tissues and organs shrink and swell to differing extents in the various solvents used to prepare tissue for microscopic examination4, s. It cannot be assumed that different components of an organ shrink or swell to the same extent; nor can it be assumed that developing, aged or diseased tissues will undergo the same volumetric changes as normal tissues. Therefore, cell numerical density can increase while the total number of cells in an organ stays the same or possibly even decreases! In this case, ceil numerical density tells us more about differential shrinkage than actual cell number.
