The electrical activity was measured by a microelectrode and it was found that the action potential of the giant axon was abolished in Na free solns., returning when sea water was replaced. Dilution of the sea water with dextrose produced a slight increase in resting potential, but a large and reversible decrease in the height of the action potential. The height of the action potential was increased by hypertonic NaCl. It was noted that these changes in membrane potential with alteration of external Na, were of the same order of magnitude as those for the Na electrode. The conduction velocity was substantially decreased in low Na content solns., and the changes produced by diluting sea water with dextrose were found to be consistent with alterations of the Na content. Increase of K caused depression of resting and action potential, whereas decrease caused some increase. The reversal of membrane potential during action potential was explained by assuming that the permeability conditions in the active state are the reverse of those in the resting state.
