Objective: To investigate the effects of Chinese medicine prescription Jiawei Xiaoyao Powder (JWXYP) on the levels of dopamine (DA) and serotonin (5-HT) in the brain of hepatocellular carcinoma (HCC) rats with liver depression and spleen deficiency. Methods: Thirty-six SD rats were randomly divided into the control group, the HCC model group (HCC) and the flavored JWXYP group (JWXYP). A rat model of HCC with liver depression and spleen deficiency was established by combining diethylnitrosamine (DEN) with chronic unpredictable mild stimulation (CUMS). The symptoms and signs of the rats in each group were observed, the pathological changes of the rats' liver were observed by HE staining, and the DA and 5-HT levels in the frontal cortex were detected by HPLC. Results:Compared with the control group, the pathological manifestations of the HCC group were hepatic lobular structure destruction, dysplastic manifestations of proliferative cells, pathological mitotic signs of binuclear cells,etc, and symptom scores were consistent with liver depression and spleen deficiency syndrome, with a significant decrease in intracranial DA and 5-HT. After JWXYP treatment, the hepatic histopathological malignancies of the rats were mild, the symptoms were significantly improved, and the cerebral DA and 5-HT were significantly increased. Conclusion: JWXYP can improve the symptoms of liver stagnation and spleen deficiency in rats, and the mechanism may be related to the increase of monoamine neurotransmitters in the brain.
