A novel method is described for the biotinylation of Ig. The procedure relies on the generation of reactive aldehydes on the carbohydrate moieties of the Ig by oxidation with sodium periodate and subsequent reaction with biotin hydrazide. The method is simple and specific and results in stable conjugates retaining full immunologic activity. It was applied successfully to several mouse monoclonal IgG and IgM antibodies and to human IgM preparations. The procedure may also be applied to conjugation of Ig with fluorescent dyes.
