The chemical structure of the cord factor of M. tuberculosis is trehalose-6,6[image]-dimycolate (1). C186H366O17 [plus or minus] 10CH2. Alkaline hydrolysis of the cord factor yields 2 molecules of mycolic acid and one molecule of the disaccharide trehalose. Methylation studies show that the 2 mycolic acid molecules are attached through an ester linkage to the primary hydroxyl groups of trehalose. Identical results were obtained with cord factor from the human strain Brevannes and with that of the BCG strain.
