Slices from rat renal cortex accumulated gentamicin (GM) against a concentration gradient. At least part of this accumulation appeared to be due to active transport as evidenced by dependence on oxygen. Classical inhibitors (both competitive and noncompetitive) of organic acid and base transport in the kidney did not reduce in vitro GM tissue to medium equilibrium ratios suggesting that GM is not transported by classical renal organic ion transport mechanisms. High concentrations of GM in vitro inhibited renal transport of the organic anion p-aminohippurate and the organic cation N1-methylnicotinamide as well as the rate of tissue respiration.
