MATERIALS AND METHODSWe used large white pigs. Three experimental groups were studied. We compared two different cold storage solutions, Euro–Collins (EC) and University of Wisconsin (UW) solutions. In the kidney preserved groups, kidneys were removed and ffushed with 500 mL of 4 C preservation solution. The kidney was wrapped in a sterile plastic bag containing 500 mL of preservation solution and stored for 48 hours on melting ice. Experimental groups were compared to a control group (uninephrectomized). Renal function was determined by creatinine clearance before surgery and after transplantation on days 1, 3, 5, 7, 11, and 14 (D1 to D14). Blood and urine were also collected for proton nuclear magnetic resonance (NMR) spectroscopy analysis until the deaths or sacrifice of the animals. For urine NMR spectra, the ratio of trimethylamine-N-oxide (TMAO) to creatinine was calculated and expressed in mmol/mol of creatinine. All values reported are means SEM and were compared by using variance analysis and a Student–Neuman Keuls test for multiple comparison.
