RESULTSRequirements for Ca++ Binding bg Xitochondria-Data shown in Table I indicate that rat kidney mitochondria can take up large amounts of Ca++ from the suspending medium. The amount of Ca++ bound was maximal when the medium contained a respiratory substrate, ATP, Pi, and Mg’+. When ATP was omitted from the optimal medium, no significant amount of Ca++ was bound (Table I, Experiment 2), whereas when Pi was omitted, the amount of Ca++ taken up was decreased from 40 to70%. The effect of Pi concentration on Ca++ binding is shown in Fig. 1; approximately 4 my Pi supported maximal Ca++ uptake. The data in Fig. 2 show the effect of STP concentration; approximately 3 rnM ATP was required for maximal Ca++ binding.
