Materials and Methods.-The mouse tumors used in these studies were the L# 2 lymphoma, 6C3HED lymphoma, and TA3 adenocarcinoma. The L# 2 was induced in an A-strain mouse by irradiation in 1946, 13 and the 6C3HED in a C3H mouse by estradiol benzoate in 1941.9 The TA3 arose spontaneously as a mammary tumor in an A-strain female.'2 Ascites sublines of these tumors were obtainedthrough the generosity of 1) r. TS Hauschka at Rloswell Park IMemorial Institute, Buffalo, NY Tumor cells were withdrawn from the peritoneal cavity, suspended in calcium-free phosphate-buffered saline (PBS), spun down at 100 X gfor 10 min, and then re-suspended in 5% EDTA and PBS.Cells from normal mouse spleen, thymus, and lung were separated from connective tissue by pressing fragments of aseptically removedorgans through a fine screen, after which they were (entrifilged and resuspended as above. Liver cells were prepared in the same way after perfusion of the liver in situ with 5%/c, EDTA. Epidermal and bone marrow suspensions were made according to Billingham's methods, 6 and erythrocytes were collected by bleeding a decapitated mouse into a tube containing 5% Oedta.
