Photosynthetic efficiency in cyanobacteria is improved under conditions of inorganic carbon (Ci) limitation by the induction of a CO2-concentrating mechanism (CCM) that elevates the CO2 concentration around the primary carboxylase ribulose-1,5-bisphosphate carboxylaseoxygenase (Rubisco) in the carboxysome. The molecular details of low-Ci sensing in relation to regulation of CCM induction in cyanobacteria are not presently known. We have applied a quantitative reverse transcription polymerase chain reaction technique to monitor the abundance of key CCM-related transcripts in Synechococcus elongatus under a variety of experimental conditions with the aim of probing the conditions required for CCM induction.
