A cowpea strain of TMV [tobacco mosaic virus] engenders rods of several sizes in infected tissues [Vigna sinensis, Phaseolus vulgaris, Nicotiana tabacum], including a short rod which contains the capsid protein gene. When full length rods were incubated at 60.degree. C with sodium dodecyl sulfate, each partially disrupted virion had an RNA strand visible at 1 end only. This partially stripped virus (PSV) was treated with RNases and then with detergent and phenol to recover the portion of the RNA protected by the unstripped protein (PROT-RNA). PSV, long rod RNA, short rod RNA and RNA (I2-RNA) from a naturally occurring rod of intermediate length, but not PROT-RNA, were enzymatically aminoacylated with valine. The long rods of this cowpea strain of TMV, like those of the common strain, are apparently stripped from the 3''-end of the RNA; the PROT-RNA must be derived from the 5''-end. PSV, long rod RNA and short rod RNA, but not PROT-RNA, completed with labeled short rod RNA in molecular hybridization reactions with melted double-stranded RNA from cowpea strain TMV-infected tissue. I2-RNA previously was shown to contain nucleotide sequences of the short rod RNA. Thus, the short and intermediate rod RNA may be derived from those long rod RNA nucleotide sequences which are close to the 3''-terminus. PROT-RNA of apparent MW 1.4 .times. 106 and long rod RNA both directed the synthesis of polypeptides of apparent MW 130,000 [in a wheat germ cell free system]. The results are consistent with the gene for this high MW polypeptide being close to the 5''-terminus of the long rod RNA, the capsid protein gene being close to the 3''-terminus, and the I2-RNA translocation product (MW .apprx. 29,000) being between.
