The relationship between the initial cleavage by restriction endonucleases and the subsequent breakdown of the cleaved fragments by exonuclease V (which is specifiied genetically by rec BC) in vivo is clarified. Using rec BC- bacteria [Escherichia coli], degradation of the fragments cleaved from the unmodified phage T4 or phage .lambda. chromosome by the cellular restriction endonucleases was prevented. Therefore these large fragments are apparently intermediates in the restriction in vivo, as postulated after in vitro experiments. Also, different restriction endonucleases which are thought to be localized in the cytoplasm or in the membrane can be inactivated in different ways by the infecting phage.
